NAD+ Not a peptide

A molecule every cell uses for energy, discussed for ageing.

StatusNot an approved medicine for these uses; precursors sold as supplements in some markets

iNot medical advice. We don’t recommend any dose. Every dose below is a quote from the person named, describing their views or their own use. NAD+ is not a peptide. NAD+ precursors such as nicotinamide riboside are sold as food supplements in some markets. We found no EU, UK or US approval of injected or intravenous NAD+ for anti-ageing or wellness uses.

What experts say about dosing

Dr Craig KoniverPhysician; founder of Koniver Wellness (Charleston, US, and London)

With patients: 100 mg under the skin, five days on

Dose
“100 milligrams”
How often
“five days on, two days off”
How taken
“subcutaneously”

“Actually, that's worked out really well. We'll do 100 milligrams subcutaneously, again, five days on, take two days off. You get a little bit of that stomach cramping from the 100 milligram injection.”

Context: What his clinics started doing during the pandemic. Before this he describes the in-clinic IV protocol they settled on: a 750 milligram “loading dose” given as five treatments in 10 days, then usually a monthly IV maintenance dose.

Own use: injections

Dose
“20 milligrams … or uh 40 milligrams split bilaterally”
How taken
“subq”

“So I, I just got sick of it. I just inject my glutathione intramuscularly three times a week 600 milligrams and NAD plus 20 milligrams subq or uh 40 milligrams split bilaterally as part of my interworkout protocol”

Own use: subcutaneous instead of NMN

Dose
“20 milligs”
How often
“per day”
How taken
“subq”

“So I, I take it subq man. I stopped nicotine monucleotides because NAD plus 20 milligs per day is subq is way way way way cheaper than 800 to 1,000 milligrams NMN.”

Dose
“100mg”
How often
“2–3 days per week”
How long
“As needed”
How taken
“SubQ injection”
When
“AM”

“Dose: 100mg. Timing: AM. Frequency: 2–3 days per week. Duration: As needed. Administration: SubQ injection. […] Jay’s Note: NAD+ isn’t a skincare compound — it’s a cellular energy compound. But when your cells have the fuel they need to repair and replicate properly, your skin reflects that.”

What experts say about safety

4 statements from 3 people about risks and how strong the evidence is.

Own use: precursor choice

“Personally, I use Nicotinamide Riboside (NR) because it is more cost effective right now, and it has human data to back up its efficacy and safety profile.”

Context: About NAD+ precursors (NR versus NMN), not NAD+ itself. Later in the article: “I take Resveratrol, a NAD+ precursor (NR as opposed to NMN), Vitamin K2 and Vitamin D.” No dose given.

Injection route and pro-oxidant effects

“um, well I think NAD plus should be done intervenous, uh, but I think you can do it subq, right […] now the problem is with injectable gluathione and N plus and other injectable antioxidants basically is that they might have some Pro, um, prooxidant effects at the site of administration”

Context: Answering a viewer who asked whether subcutaneous NAD+ is worth it. Captions render NAD+ as “N plus” and glutathione as “gluathione”. The cut removes a remark about grey-market products.

Own experience: NR vs NMN

“Every time I took NR up to 1,200 milligs per day, I got a minute effect compared to a low dose of NMN. Now I've supplemented between 250 to 1,000 milligs of NMN per day. That's either with NAD+ administrations or without it.”

Side effects of higher doses

“And going higher doesn't get you more. It just gets you closer to more side effects, like the flushing of the face, the tightness in the chest”

What the research used

Context, not a dosing guide. Human data on injected NAD+ are very limited. Most trials test oral precursors (NR, NMN) rather than NAD+ itself. None of this establishes an injection dose.

Human studies

StudyStudied inWhat the study usedWhat it found
Grant et al., 2019, Front Aging NeurosciPubMed 31572171Healthy men (small pilot)IV NAD+ at 3 µmol/min for 6 hoursNo change in plasma NAD+ for the first 2 hours; metabolites appeared in urine later
Christen et al., 2026, Nat MetabPubMed 4154025365 healthy adultsOral NR, NMN or nicotinamide for 14 daysNR and NMN raised blood NAD+ to a similar degree; nicotinamide did not
What we don’t know yet
  • NAD+ is a coenzyme, not a peptide.
  • Very few human studies of injected NAD+.
  • Flushing and chest tightness are mentioned by Nicholas Trigili as side effects of higher doses; we found no trial quantifying them.
  • Grey-market products are unregulated, so identity, purity and dose accuracy vary.
Common questions

Is NAD+ a peptide?

No. It is a coenzyme made from vitamin B3. We list it because it is often used in peptide stacks.

Does oral NR or NMN raise NAD+?

In a 2026 trial in 65 healthy adults, both raised blood NAD+ over 14 days.

Why does Nicholas Trigili’s dose say micrograms?

That is the unit he says, twice, in the video. We have not changed it.

Why does the protocol section only contain quotes?

We do not write our own dose ranges. The protocol cards show, word for word, what named people have publicly said, with their caveats and a link to the source.

About this page

Word-for-word statements about injectable NAD+ and NAD+ precursors, each with a link to the source. NAD+ is a coenzyme, not a peptide; it is included because it is often used alongside peptides. The research is listed separately as context.
