Own use: how 25 mg compares

Dose
“25 mg”

“If I had to compare 25 mg of MK-677 to something I would compare the results to 2 iu’s of high quality generic GH, or a few daily 100 mcg injections of GHRP-6 coupled with concurrent 100 mcg injections of Modified GRF (1-29).”

Context: From the “My MK-677 Review” section, where he says “I have used MK-677 a few times over the past couple years now.” A comparison of effects, not a recommendation.

10 mg vs 25 mg

Dose
“10 mg … 25 mg is the benchmark”

“Based on the study, if fat loss and the anti-aging/healing benefits of HGH is all that is sought after by the user, a 10 mg dose would likely be sufficient. Whereas if the user was seeking maximized muscle growth potential, then higher IGF-1 levels would be what they are after, and would then justify a higher dosing protocol. […] Hence, 50 mg is unnecessarily high to research with, and 25 mg is the benchmark for boosting IGF-1 levels significantly higher, while still reaping all of the benefits of the GH increase.”

Harm-reduction note (he says he will never recommend it)

Dose
“anywhere from 10 to 25 milligs”
How often
“a day”
How long
“cycle it; not indefinitely”

“I can't and will never recommend the use of a research chemical to anyone. But in the spirit of harm reduction, if you insist on trying it, you must be smart. Monitor your blood sugar, do your cardio, watch your blood pressure, keep the dose reasonable, with most research using anywhere from 10 to 25 milligs a day, and cycle it. Do not use this indefinitely.”

Context: He frames this as harm reduction, not a recommendation. The 10 to 25 range is what he says most research used. “milligs” is the caption wording for milligrams.

Derek (More Plates More Dates)Fitness educator; founder of More Plates More Dates

Own use: lethargy and appetite

“In terms of side effects, I have noticed some fairly significant lethargy from MK-677 (anything that raises GH and IGF-1 will do this), however, I was able to mitigate that to some extent by taking my daily dosage right before going to bed.”

Context: Elsewhere in the same review he says: “The incredibly heightened appetite is the main reason I avoid this compound now”.

Limit the cycle length

“if you take it you have to be very controlled in the timeframe that you take it, like I wouldn’t recommend more than 62 possibly 12 weeks of a cycle with something like that. And then you have to go off of it for a while, because you’re going to change that receptor and it’s going to involute”

Context: He is warning that strong ghrelin-receptor drugs such as MK-677 can switch the receptor off, which he links to anxiety and low mood. “62” is how the published transcript renders it; the spoken words are probably “6 to”. He gives no dose.

Tier list: F tier

“So, MK677 is going to be an F tier, mainly because you can't get a prescription MK677 anymore, and it raises growth hormone and IGF-1, which is great, but then drawbacks is it wrecks your hunger. It increases grein, which you're going to be hungry all the time. You're not going to be able to control your appetite. This is not good for most people. A lot of water retention, a lot of inflammation, and you can't control your glucose.”

Context: “grein” is how the transcript renders ghrelin. He gives no dose.

Blood sugar and insulin sensitivity

“It also raised fasting blood glucose by roughly 5 mg/dL and reduced insulin sensitivity. Appetite went up, as you would expect from agonizing the hunger receptor. Transient lower limb edema and muscle pain were the other common complaints.”

What the research used

Context, not a dosing guide. MK-677 has been tested in randomised trials, mostly in older adults, at 2–25 mg a day. These were medically supervised studies, so none of this establishes a dose for anyone else.

Human trials

StudyStudied inWhat the study usedWhat it found
Chapman et al., 1996, J Clin Endocrinol MetabPubMed 895402332 healthy adults aged 64–812, 10 or 25 mg by mouth once daily for 2 and 4 weeksGH and IGF-1 rose with dose; 25 mg/day raised 24-hour GH by 97%
Nass et al., 2008, Ann Intern MedPubMed 1898148565 healthy adults aged 60–81, two years25 mg by mouth once daily vs placeboFat-free mass rose 1.1 kg vs a 0.5 kg fall on placebo; fasting glucose rose ~5 mg/dL and insulin sensitivity fell; no change in strength or function
What we don’t know yet
  • The long trials were in older adults; there are no long-term trials in young, healthy people.
  • Raises appetite and blood sugar and lowers insulin sensitivity in trials.
  • Grey-market products are unregulated, so identity, purity and dose accuracy vary.
Common questions

Is MK-677 a peptide?

No. It is a small molecule taken by mouth. It is listed here because it is discussed alongside the growth-hormone-releasing peptides and acts on the same receptor.

Why do people take it at night?

Vigorous Steve’s range is “once daily before bed”, and Derek says taking it before bed reduced the lethargy he felt.

What are the common side effects?

In the two-year trial: more appetite, mild swelling of the lower legs, muscle pain, higher fasting blood sugar and lower insulin sensitivity. Derek and Nicholas Trigili describe strong hunger.

Why does the protocol section only contain quotes?

We do not write our own dose ranges. The protocol cards show, word for word, what named people have publicly said, with their caveats and a link to the source.
